Test Details

Methodology

Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry is the primary identification method. Ancillary testing (ie, 16S gene sequencing, ITS gene sequencing, conventional biochemicals) will be employed as required.

Result Turnaround Time

5 - 15 days

Turnaround time is defined as the usual number of days from the date of pickup of a specimen for testing to when the result is released to the ordering provider. In some cases, additional time should be allowed for additional confirmatory or additional reflex tests. Testing schedules may vary.

Use

Determine the identification of isolates (ie, bacterial, mycobacterial, etc) from another laboratory that has performed culture of the clinical specimen. These isolates may include strict aerobes, facultative anaerobes, anaerobes, yeast, mycobacteria, Nocardia and/or Actinomycetes.

Special Instructions

Susceptibility testing may be requested for an additional charge. The client submitting the isolate for testing should provide Labcorp with clinical and testing information regarding test procedures already performed at the submitting laboratory in order to expedite testing on receipt.

Broth medium containing a pure culture may be submitted for testing; however, additional time will be required in order to perform subculture to solid media prior to initiation of MALDI testing. Testing will reflex to sequencing when ID cannot be determined by MALDI-TOF.

The technology is limited to what is claimed in the spectral database. If organism is not obtained, the test will reflex to identification by ITS gene sequencing.

Specimen Requirements

Specimen

Pure culture isolate of bacteria, yeast, filamentous fungi, AFB, Nocardia, or aerobic Actinomycetes

Volume

Pure culture (visible colonies should be present). Bacterial culture transport swab with media inoculated with the isolate from a pure culture or agar slant in a screw-cap container packed as an etiologic agent. Maintain a subculture at the submitting laboratory.

Minimum Volume

Viable colonies macroscopically visible on culture medium; one bacterial culture transport swab with media.

Bacterial swab transport inoculated with the isolate from a pure culture or agar slant in screw-cap container packed as an etiologic agent. Maintain a subculture at the submitting laboratory.

Isolate nonviable; unlabeled specimen or name discrepancy between the specimen and the name in the computer or on the test request form; isolate transported under inappropriate conditions; broken tube or plate; mixed culture; single specimen identifier
